Measurement of intracellular Ca2+ concentration [Ca2+]i. a Time course of intracellular Ca2+ levels in WT- or pGly291Ser-expressing HEK293T cells. An increase in Ca2+ level after application of 30 mM Ca2+, an indication of plasma membrane Ca2+ permeability, was observed for WT cells. In cells expressing p.Gly291Ser, the Ca2+ level was significantly higher than that for WT in the steady state (2 mM Ca2+, time = 0; p = 0.012 vs. WT, n = 39–49, Mann–Whitney rank-sum test), and the level did not increase substantially upon application of 30 mM Ca2+, most likely due to saturation. These results suggest a Ca2+ overload in the steady state
