A proposed working model showing Piezo1-mediated suppression of differentiation in brown adipocytes. A Piezo1 agonist (Yoda-1) and endogenous stimuli including mechanical stimuli activate Piezo1, followed by an increase in the [Ca2+]i in pre-adipocytes and the early stage of differentiation in brown adipocytes. Ca2+ binds to calmodulin (CaM), and the Ca2+/CaM complex activates calcineurin, which suppress the expression of the key transcriptional factors PPARγ and C/EBPα, which in turn suppress the differentiation of brown adipocytes
